Clarence Road from its junction with Brighton Road to Cambridge Road junction will be shut from May 16-18.
West Sussex County Council says the closure is necessary for Southern Water to undertake works to a leaking pipe.
It says that access will be maintained for emergency services, residents and pedestrians.
